Abstract
Los oligodendrocitos son las células mielinizantes del sistema nervioso central. Se ha demostrado la presencia de CB1 en áreas específicas de la sustancia blanca, OPCs del nervio óptico y células A2B5+ in vivo (Herkenham y cols., 1991; Molina-Holgado y cols., 2002; Mato et al, 2009;. Sim y cols., 2006). Por otra parte, los agonistas cannabinoides aumentan la expresión de la proteína básica de mielina en la sustancia blanca subcortical del cerebro de rata neonatal (Arevalo-Martin y cols., 2007). Los endocannabinoides regulan la neurogénesis controlando la proliferación, migración y diferenciación de células madre / precursores neurales (NSPC) (Aguado y cols., 2005; Palazuelos y cols., 2006; Molina-Holgado y cols., 2007; Galve-Roperth y cols., 2013). Existen pocos pero importantes datos sobre la acción de los endocannabinoides en los oligodendrocitos. En esta tesis nuestro primer objetivo fue estudiar in vitro la presencia de algunos de los componentes del sistema endocannabinoide en los OPCs, incluyendo 2-araquidonilglicerol (2- AG), CB1, CB2, DAGLα y β (las principales enzimas responsables de la síntesis del 2-AG) y monoacilglicerol lipasa (MAGL), responsable de la hidrólisis de este endocannabinoide (Dinh y cols., 2002). A continuación, se analizó el efecto del 2-AG en tres de las funciones más importantes en el desarrollo de los oligodendrocitos: proliferación, migración y diferenciación. Oligodendrocytes are the myelinating cells of the central nervous system and the presence of cannabinoid CB1 receptors have been observed in specific white matter areas, in OPCs from optic nerve and also in A2B5 positive-OPCs obtained from human brain (Herkenham et al., 1991; Molina-Holgado et al., 2002; Mato et al., 2009; Sim et al., 2006). Moreover, in the subcortical white matter of neonatal rat brain cannabinoid agonists enhance the expression of myelin basic protein (Arevalo-Martin et al., 2007) and endocannabinoids regulate neurogenesis controlling proliferation, migration and differentiation of neural stem/precursor cells (NSPC) (Aguado et al., 2005; Palazuelos et al., 2006; Molina-Holgado et al., 2007; Galve-Roperth et al., 2013). Therefore, there is limited data about the action of endocannabinoids in oligodendrocyte cells. In this thesis our first objective was to study in vitro the presence of some of the components of the endocannabinoid system in OPCs, including 2-araquidonoyl glycerol (2-AG), CB1, CB2, DAGLα and β (the main enzymes responsible of the synthesis of 2-AG and monoacylglycerol lipase (MAGL), responsible of the hydrolysis of this endocannabinoid (Dinh et al., 2002). Next we analyzed the effects of 2-AG on three key functions in oligodendrocyte development: proliferation, migration and differentiation. Finally, we studied some of the signaling pathways underlying these processes. Our results demonstrated that oligodendrocyte cells express not only the cannabinoid receptors, but also the enzymatic machinery for the synthesis and degradation of the endogenous ligand 2-AG, which acts in an autocrine or/and paracrine manner (Gómez et al., 2010). A2B5 positive OPCs strongly expressed both DAGLα and β and although MBP positive oligodendrocytes still expressed both enzymes it seemed to be less abundant. On the contrary, MAGL expression is markedly higher in mature oligodendrocytes compared with OPCs. Next, the analysis of lipid extracts from developing oligodendrocyte cultures by liquid chromatography coupled to mass spectrometry demonstrated that oligodendrocyte cells produce 2-AG and there is a variation in 2-AG concentration that relates to the expression of DAGLα and β and MAGL; the total amount of 2-AG in OPCs was significantly higher than in mature oligodendrocytes...
