Abstract
Los receptores purinérgicos P2X7 están implicados tanto en procesos fisiológicos como es el crecimiento y ramificación axonal o neuroprotección. Pero además está implicado en procesos patológicos como epilepsia, Huntington o Alzheimer. Sin embargo se conoce muy poco sobre su regulación transcripcional. En esta tesis se describe por primera vez la implicación del factor de transcripción Sp1 en la regulación de la expresión del receptor P2X7. Para ello se ha delimitado una región mínima en el promotor del gen P2rx7 de ratón que contiene cuatro sitios SP1, dos de ellos conservados en mamíferos. La funcionalidad de estos sitios SP1 se confirmó por mutagénesis dirigida y sobreexpresión e interferencia de Sp1 en células de neuroblastoma de ratón Neuro2a. Además, el tratamiento farmacológico con mitramicina A, redujo los niveles endógenos de expresión del receptor en cultivos primarios de neuronas y astrocitos de corteza de ratón. El uso del ratón transgénico P2xr7-EGFP que expresa la proteína verde fluorescente bajo el promotor de P2xr7 nos permitió observar una elevada correlación entre células que expresaban Sp1 y GFP en el cerebro, demostrando que Sp1 es un elemento clave en la regulación transcripcional del receptor P2X7 en el sistema nervioso. Trabajos previos indicaban que la ausencia de suero puede activar el receptor del factor de crecimiento epidérmico (EGFR) al alterarse la estructura de la bicapa lipídica. La inhibición farmacológica del EGFR nos permitió identificar su implicación en la regulación de la transcripción del receptor P2X7. Además, la activación del EGFR inicia una cascada de señalización a través de PI3K y Akt capaz de potenciar la actividad de Sp1, facilitando su entrada a núcleo y favoreciendo la transcripción de P2rx7. Por otra parte, la retirada de suero provoca la activación de PKCζ, una PKC atípica que parece estar regulando negativamente la expresión de P2X7. P2X7 receptors are involved not only in physiological as axonal branching and neuroprotection but also in pathological brain processes such as epilepsy, Alzheimer disease or Huntington. However little is known about its transcriptional regulation. In my PhD studies we reported the first molecular and functional evidence that Specificity protein 1 (Sp1) transcription factor plays a pivotal role in the transcriptional regulation of P2X7 receptor. We delimited a minimal region in the murine P2rx7 promoter containing four SP1 sites, two of them being highly conserved in mammals. The functionality of these SP1 sites was confirmed by site-directed mutagenesis and Sp1 overexpression/ down-regulation in the neuroblastoma cell line Neuro2a. Inhibition of Sp1-mediated transcriptional activation by mithramycin A reduced endogenous P2X7 receptor levels in primary cultures of cortical neurons and astrocytes. Using P2rx7-EGFP transgenic mice that express enhanced green fluorescent protein under the control of P2rx7 promoter, we found a high correlation between GFP and Sp1 levels in the brain, demonstrating that Sp1 is a key element in the transcriptional regulation of P2X7 receptor in the nervous system. Previous works reported that serum starvation could activate epidermal growth factor receptor (EGFR) due to the disruption of the lipid bilayer. The pharmacological inhibition of the EGFR, showed its involvement in P2X7 transcriptional regulation. Besides, EGFR activation initiates a signaling pathway through PI3K and Akt able to enhance Sp1 activity favoring P2xr7 transcription. On the other hand, serum withdrawal evokes PKCζ activation, which seems to be negatively regulating P2X7 expression.
